摘要: Mu DNA transposition proceeds through a series of higher-order nucleoprotein complexes called transpososomes. The structural core the transpososome is tetramer transposase, A, bound to two transposon ends. High-resolution analysis intact transposase and has not been successful date. Here we report structure A at 16-A Type 1 34-A resolution, by 3D reconstruction images obtained scanning transmission electron microscopy (STEM) cryo-temperatures. Electron spectroscopic imaging (ESI) DNA-phosphorus was performed in conjunction with investigation derive path define DNA-binding surface transposase. Our model fits well accumulated biochemical literature for this intricate system, lays foundation function, including catalysis trans complex circuit macromolecular interactions underlying transposition.
