Cathepsin B-cleavable doxorubicin prodrugs for targeted cancer therapy (Review)

摘要: Doxorubicin (DOX) is one of the most effective cytotoxic anticancer drugs used for treatment hematological malignancies, as well a broad range solid tumors. However, clinical applications this drug have long been limited due to its severe dose-dependent toxicities. Therefore, DOX derivatives and analogs developed address issue. A type prodrug, cleaved by cathepsin B (Cat B), which highly upregulated in malignant tumors premalignant lesions, has achieve higher concentration tumor tissue lower normal tissue, so enhance efficacy reduce toxicity cells. In review, we focused on Cat B-cleavable prodrugs discussed these prodrugs, demonstrated preclinical developments.
