摘要: For the potential use of Wheat germ agglutinin (WGA) and Solanum tuberosum lectin (STL) as auxiliary excipients for targeting drugs to colonocytes, number Caco-2 HT-29-bound lectins was determined by fluorimetry using fluorescein-labelled derivatives N-acetylglucosamine-specific lectins. After 1 h incubation, WGA-binding capacity 5 x 10(4) cells 26.9 +/- 0.5 pmol at 4 degrees C 27.2 1.0 37 respectively. In comparison, 19.5 2.9 (37 C) 16.7 0.9 (4 WGA were bound within HT-29 referring about 80% total amount incubation. contrast, binding STL colon carcinoma cell lines independent incubation times temperatures tested exhibiting a rate 8.4 0.6 (HT-29) 9.9 0.8 (Caco-2) STL/5 cells. As flow cytometry, non-specific is lower than 1.0% 3.4% (STL). Uptake intracellular accumulation investigated confocal laser scanning microscopy A decrease initially membrane-bound concurrent with increasing cytoplasmic enrichment time observed digital image analysis. Due specific numerically sufficient adhesion well internalization, are anticipated tools in lectin-mediated drug delivery systems.
