摘要: Tumor-associated lymphocytes (TAL) were isolated from the ascitic fluid of patients with adenocarcinoma ovary. These cells proliferated and expanded by 100-600-fold as either CD3+ CD4+ or CD8+ cultures in presence moderate concentrations (50-200 cetus units/ml) recombinant interleukin 2 reached high numbers (5 x 10(8)-1 10(9)). After expansion 16 TAL samples 15 patients, 5 7 ovarian cytotoxic T-lymphocyte cell lines T-cell receptor (TCR) (alpha beta)+ CD4- phenotype exhibited preferential cytolytic activity against autologous tumor targets significantly lower allogeneic natural killer-sensitive line K562. The was inhibited operationally anti-TCR beta) monoclonal antibody specific for CD3 differentiation antigen, indicating that TCR are involved process. other demonstrated similar both tumors. these predominantly CD8-. Certain clones representative tumor-specific cytotoxicity may be major histocompatibility complex restricted. Other nonmajor restricted cytotoxicity. results demonstrate T-cells present malignant ascites can propagated large long time intervals vitro. This finding significant further investigation T-lymphocytes future adoptive immunotherapy studies.
