Endostatin Gene Transfer Inhibits Joint Angiogenesis and Pannus Formation in Inflammatory Arthritis

摘要: Rheumatoid arthritis is a prevalent example of an inflammatory angiogenic disease, which mediated by pro-inflammatory and pro-angiogenic cytokines such as tumor necrosis factor (TNF). To evaluate the effect potent anti-angiogenic endostatin on TNF-induced arthritis, we injected endostatin-expressing lentiviral vector directly into joints human TNF-transgenic mice before onset disease. Histological analysis 8 weeks later revealed that reduced blood vessel density within synovial tissues overall mean index. In vitro in vivo examination potential mechanism inhibited blocks activation JNK JNK-dependent gene expression. These data suggest novel inhibits angiogenesis, demonstrates utility therapy for treatment arthritis.
