Cross-Repressive Interaction of the Olig2 and Nkx2.2 Transcription Factors in Developing Neural Tube Associated with Formation of a Specific Physical Complex

摘要: In developing neural tube, the basic helix-loop-helix (bHLH) transcription factor Olig2 interacts with homeodomain Nkx2.2 at two distinct stages. During neuronogenesis, a cross-repressive interaction appears to establish precise boundary between p3 and pMN domains. At later times, cooperative is noted because promotes maturation of oligodendrocyte progenitor cells specified by expression Olig2. We show here that protein can form physical complex in mammalian yeast two-hybrid trap assay. This specific does not bind biologically irrelevant (Nkx6.1), interact bHLH (NeuroD). Deletion mapping analysis suggests formation an Olig2-Nkx2.2 insufficient for induction progenitors spine; however, protein-protein observed might be important helps pMN-p3 spinal cord.
