摘要: A new cobalt(II) complex (1) of 5-chloro-8-hydroxyquinoline was prepared and structurally characterized using infrared spectroscopy, electrospray ionization mass spectrometry, elemental analysis, single-crystal X-ray diffraction as well powder diffraction. Its biological activities including DNA binding anticancer activity were investigated. The study 1 suggested that it interacted with calf thymus mainly via an intercalative mode. in vitro screened against a series tumor cell lines the normal liver line HL-7702 MTT assay. showed much higher cytotoxicity than corresponding metal salt ligand towards five tested lines, which T-24 most sensitive 1, IC50 value 7.04 ± 0.06 μM. found to greatly induce cycle arrest cells at S phase, consequently apoptosis dose-dependent mode by analysis Hoechst 33258 acridine orange/ethidium bromide staining assays. mechanism studied targeting mitochondrion-mediated pathway, since apoptotic treated investigated reactive oxygen species (ROS) detection, intracellular calcium concentration measurement caspase-9/3 results induced closely related loss mitochondrial membrane potential, ROS production enhancement [Ca2+], would trigger activation dysfunction pathway.
