摘要: S-layer fusion protein technology was used to design four different fluorescent proteins with three GFP mutants and the red mRFP1. Their absorption spectra, steady-state fluorescence, fluorescence lifetime were investigated as a function of pH. It found that intensities mutant decreased about 50% between pH 6 5. The spectral properties minimally affected by variations. These results compared His-tagged reference proteins, demonstrating did not change general whole protein. In addition, pKa values calculated.
