Tri-ortho-cresyl phosphate induces autophagy of rat spermatogonial stem cells

摘要: Tri-ortho-cresyl phosphate (TOCP) has been widely used as plasticizers, plastic softeners, and flame retardants in industry reported to have a deleterious effect on the male reproductive system animals besides delayed neurotoxicity. Our preliminary results found that TOCP could disrupt seminiferous epithelium testis inhibit spermatogenesis, but precise mechanism is yet be elucidated. This study shows inhibited viability of rat spermatogonial stem cells dose-dependent manner. not lead cell cycle arrest cells; mRNA levels p21, p27, p53, cyclin D1 were also affected by TOCP. Meanwhile, did induce apoptosis cells. After treatment with TOCP, however, both LC3-II ratio LC3-II/LC3-I markedly increased; autophagy proteins ATG5 beclin 1 increased after indicating Ultrastructural observation under transmission electron microscopy indicated autophagic vesicles cytoplasm containing extensively degraded organelles such mitochondria endoplasmic reticulum significantly treated In summary, we shown can cells, without affecting apoptosis.
