摘要: Previous studies have demonstrated that allelic deletions of the short arm chromosome 17 occur in over 75% colorectal carcinomas. Twenty 17p markers were used to localize common region deletion these tumors a contained within bands 17p12 17p13.3. This contains gene for transformation-associated protein p53. Southern and Northern blot hybridization experiments provided no evidence gross alterations p53 or surrounding sequences. As more rigorous test possibility was target deletions, coding regions from two analyzed; tumors, like most carcinomas, had expressed considerable amounts messenger RNA remaining allele. The allele mutated both with an alanine substituted valine at codon 143 one tumor histidine arginine 175 second tumor. Both mutations occurred highly conserved previously found be murine oncogenes. data suggest may involved neoplasia, perhaps through inactivation suppressor function wild-type gene.
