Patterns of dna amplification at band q13 of chromosome 11 in human breast cancer

摘要: In an attempt to verify the nature of amplification events at band q13 on chromosome 11 we surveyed status ten molecular markers specific for this region (GSTP, SEA, D11S97, D11S146, BCLI, PRAD1/CCND1, HST/FGF4, INT2/FGF3, EMS1, and D11S833E) in a panel 389 primary breast carcinoma DNA samples. Eighty-eight tumors (23%) showed least one these amplified, but majority cases encompassed more than tested loci. Our data confirm that amplicons 11q13 can cover large portions are consistent with existence several cores amplification. One important core seems be, as previously described, centered around PRAD1/CCND1; 57 (14.7%) PRAD1/CCND1 either alone (one tumor) or along BCL1 INT2/FGF3. The level sometimes exceeded surrounding markers. Three additional occurring independently were also detected. Centromeric BCL1, probes D11S146 detected 60 (15.4%) often only amplified Telomeric D11S833E was found tumors, it most marker another six cases. At shorter distance EMS1 two could exceed D11S833E. thus suggest four independent regions within cancer. Genes Chrom Cancer 9:42-48 (1994). © 1994 Wiley-Liss, Inc.
