摘要: The renin angiotensin system (RAS) has been implicated as one mediator of the cardiovascular effects estrogen. Since changes in type 1 (AT1) receptor expression are central to modulation RAS, we used noninvasive PET imaging technique study for vivo estrogen on membrane and intracellular AT1 receptors. Methods: Dynamic measurements canine (cAT1) receptors using radiolabeled antagonist, 11 C-L-159,884, were performed during 2-wk consecutive periods deprivation induced by ovariectomy 17-estradiol (E2) replacement. Results: Kinetic modeling time–activity curves kidney adrenal showed lower replete state (21% 30% decrease Gjedde– Patlak slope, influx constant, respectively). These findings correlated with vitro radioligand-binding assays 125 I-[Sar ,Ile 8 ]angiotensin II showing reduced number (35%), glomeruli (30%), myocardium liver (21%) estrogen-replenished compared estrogen-depleted animals.
