摘要: The GATA4, 5 and 6 subfamily of transcription factors are potent transactivators expressed within the precardiac mesoderm. However, little is known immediate downstream targets GATA-factor regulation during earliest stages cardiogenesis. Using P19-CL6 embryonal carcinoma (EC) cell line as an in vitro model cardiogenesis, we show that GATA6 most abundantly GATA presumptive cardiac cells. Consequently, performed a microarray screen comparing mRNA from control EC cells, early differentiation pathway, with those which had been overexpressed. These studies identified 103 genes whose expression changed significantly this was verified representative array these by real-time RT-PCR. We one genes, Wnt2, mirrors vivo. In addition, its upregulation differentiating cells mediated direct binding GATA-factor(s) to cognate Wnt2 promoter, suggesting target
