Downregulation of myosin VI reduced cell growth and increased apoptosis in human colorectal cancer

摘要: Colorectal cancer (CRC) is the third most commonly diagnosed worldwide, with mortality increasing steadily over last decade. Myosin VI (MYO6) expression found to be elevated in some types of human carcinoma cell types, suggesting that it may a sensitive biomarker for diagnosis and follow-up. In this study, we first used Oncomine database explore MYO6 CRC tissues, then constructed plasmid RNA interference targeting gene. After transfection lentivirus into SW1116 cells, viability proliferation were measured 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide colony formation assay. Cell cycle distribution was assayed by flow cytometry apoptosis evaluated Annexin V. detected quantitative real-time polymerase chain reaction western blot analysis. It mRNA upregulated tissues using data mining public microarray datasets. Depletion significantly inhibited formation. addition, knockdown slightly arrested G0/G1 phase, but remarkably increased proportion sub-G1 phase increase apoptotic cells. These results suggest promote growth as potential target anticancer therapy CRC.
