摘要: Despite major achievements in the understanding of human disease, there is a general perception that drug development industry has failed to meet expectations recent advances biotechnology should drive. One potential sources failure many next generation drugs their targets are embedded highly nonlinear signaling pathways and gene networks with multiple negative positive feedback loops regulation. There increasing evidence this complex network shapes response external perturbations form treatment, originating bistability, hypersensitivity, robustness, dose-response curves or schedule dependent activity. This review focuses on effect nonlinearities involved using tools from Nonlinear Dynamics discuss implications overcome effects regulatory networks.
