Superovulation alters the expression of imprinted genes in the midgestation mouse placenta

摘要: Imprinted genes play important roles in embryonic growth and development as well placental function. Many imprinted acquire their epigenetic marks during oocyte growth, this period may be susceptible to disruption following hormonal stimulation. Superovulation has been shown affect of the embryo, but an effect on not postimplantation embryos. In present study, we examined superovulation/in vivo or superovulation/3.5dpc (days post-coitum) embryo transfer allelic expression Snrpn, Kcnq1ot1 H19 embryos placentas at 9.5 days gestation. followed by resulted biallelic Snrpn 9.5dpc while was affected; embryos, there normal monoallelic three genes. We did observe significant DNA methylation perturbations differentially methylated regions H19. 3.5dpc placenta. The factor closely linked H19, Insulin-like factor-II, increased placenta superovulation with without transfer. These results show that both maternally paternally were affected, suggesting compromises quality interferes maintenance imprinting preimplantation development. Our findings contribute evidence mechanisms for maintaining are less robust trophectoderm-derived tissues, have clinical implications screening patients assisted reproduction.
