Interaction between phospholipid bilayer membranes and the polyene antibiotic amphotericin B: Lipid state and cholesterol content dependence

摘要: The interaction between amphotericin B and egg yolk phosphatidylcholine, dimyristoyl (DMPC) dipalmitoyl phosphatidylcholine (DPPC) phospholipid bilayer vesicles has been monitored by the circular dichroism (CD) spectra of at a 1 . 10(-5) M concentration. This method revealed that may be present in number different forms depending on time elapsed after mixing, cholesterol content vesicles' physical state. Some striking features these CD detected species are following: with molar percentage lower than 25, 25 degrees C several coexistent, their amount is time-dependent; or phosphatidylcholines without gel state, form from former appears very rapidly; DMPC DPPC 50 new monitored, identical three cases observed liquid crystalline state as well In case phospholipids definite antibiotic but characteristics according to nature lipid. With 'Fungizone' same appearance much slower. Two explanations proposed for origin discrepancies electronic absorption.
