MHC Class I–Associated Phosphopeptides Are the Targets of Memory-like Immunity in Leukemia

摘要: Deregulation of signaling pathways is a hallmark malignant transformation. Signaling-associated phosphoproteins can be degraded to generate cancer-specific phosphopeptides that are presented by major histocompatibility complex (MHC) class I and II molecules recognized T cells; however, the contribution these phosphoprotein-specific cells immune surveillance unclear. We identified 95 on surface primary hematological tumors normal tissues, including 61 were tumor-specific. Phosphopeptides more prevalent aggressive samples. CD8(+) cell lines specific for killed both leukemia human leukocyte antigen-matched ex vivo. Notably, healthy individuals showed robust responses against many within circulating memory compartment. This immunity was significantly reduced or absent in some patients. reduction correlated with clinical outcome; restored after allogeneic stem transplantation. These results suggest may targets cancer humans, point their importance development vaccine-based adoptive transfer immunotherapies.
