SIR–nucleosome interactions: Structure–function relationships in yeast silent chromatin

摘要: Discrete regions of the eukaryotic genome assume a heritable chromatin structure that is refractory to gene expression, referred as heterochromatin or "silent" chromatin. Constitutively silent found in subtelomeric domains number species, ranging from yeast man. In addition, chromatin-dependent repression mating type loci occurs both budding and fission yeasts, enable sexual reproduction. The silencing characterized by an assembly Silent Information Regulatory (SIR) proteins-Sir2, Sir3 Sir4-with unmodified nucleosomes. Silencing requires lysine deacetylase activity Sir2, extensive contacts between nucleosome, well interactions among SIR proteins, generate Sir2-3-4 complex. Results recent structural reconstitution studies suggest updated model for ordered organization SIR-dependent yeast. Moreover, expression reveal importance regulation genes other than loci. This review covers advances this field.
