摘要: With the development of a direct visualization sex chromosome in single sperm by fluorescence situ hybridization (FISH) technique, frequency aberration (aneuploidy) spermatozoa several mammals has been investigated. However, there is no report incidence X-Y aneuploidy population dogs. Therefore, this study, dog was examined multicolor FISH using specific molecular probes for canine chromosomes and autosome. Semen from eight male Labrador retrievers used as specimen. For decondensation nuclei, specimen treated with 1 M NaOH 4 minutes at room temperature. Probes X, Y, 1, labeled SpectrumGreen, Cy3 Cy5, respectively, were hybridized decondensed spermatozoa. Fluorescence signals heads clearly detected each specimen, regardless donor. The signal least one three all examined. There significant difference between theoretical ratio (50:50) observed X Y Mean percentage 0.127% (ranged 0% 0.316%). This lower than reported cattle, horses, river buffalo, goats sperm, but higher that mice sheep.
