摘要: Restriction landmark genomic scanning (RLGS) is a novel method which enables us to simultaneously visualize large number of loci as two-dimensional gel spots. By this method, the status DNA methylation can efficiently be determined by monitoring appearance or disappearance spots using methylation-sensitive restriction enzyme. In present study, RLGS with NotI, we examined, in comparison brain profile, more than 900 among three types mouse cell lines: embryonal carcinoma line P19, stable mesenchymal 10T1/2, and our established neuroepithelial (EM) lines. We found that relative numbers appeared were less 3.3% those surveyed all lines examined. However, 5 14% disappeared, increasing an increase length culture period, many commonly lost 10T1/2 EM Thus, for these lines, disappeared appeared. disappearing appearing well balanced, ratio P19 cells was almost equal liver vivo. These experimental observations suggested permanent such are hypermethylated newly also becoming heavily methylated at common loci. On other hand, demethylation seem balanced manner similar vivo tissue.
