摘要: Cutaneous T-cell lymphoma (CTCL) is a heterogeneous group of extra-nodal non-Hodgkin lymphomas with primary manifestation in the skin poor treatment options advanced stages. As basis for future immune-therapeutic strategies we have investigated possible expression tumor-specific targets CTCL focusing mainly on so-called cancer-germline genes. cDNAs derived from 20 tissues and 4 cell lines were tested 15 gene-specific gene family-specific primers by RT-PCR confirmative Northern blotting. The most frequently detected mRNAs LAGE-1 (55% only partial coexpression splicing variants), cTAGE-1 (35%), MAGE-A9 (27%) GAGE-3–7 (35%). Furthermore, could detect NY-ESO-1 (21%) MAGE-A subgroup (15%), whereas sub-specification latter proved absence MAGE-A1, -A2, -A3, -A6 -A12. SCP-1 was found one specimen several antigens not been any tumor tissue or line (MAGE-B, GAGE-1,2,8 all RAGE genes). 90% samples positive at least 1 frequent (LAGE-1, NY-ESO-1, cTAGE-1, MAGE-A9, GAGE-3to7). Using secondary SEREX approach sero-reactivity sera patients against recombinant (10/29), GAGE (3/19), MAGE-A1 (1/18), -A3 (2/18) -A9 (4/18) protein, but LAGE-1a, MAGE-A4b MAGE-A12 protein (n = 19). We conclude, that certain genes can be are able to elicit systemic immune response. These candidate might therefore promising immunotherapeutic interventions CTCL. © 2003 Wiley-Liss, Inc.
