摘要: Cynandione A, an acetophenone isolated from Cynanchum Wilfordii Radix, attenuates inflammation. The present study aimed to the mechanisms underlying cynandione A-induced antiinflammation. Treatment with A and specific α7 nicotinic acetylcholine receptor (α7 nAChR) agonist PHA-543613 remarkably reduced overexpression of proinflammatory cytokines, including tumor necrosis factor (TNF)-α, interleukin (IL)-6 IL-1β in lipopolysaccharide (LPS)-treated RAW264.7 cells primary peritoneal macrophages, endotoxemic mice. Both also stimulated IL-10 expression naive LPS-treated macrophages A- PHA-543613-inhibited cytokine was completely blocked by nAChR antagonist methyllycaconitine antibody. stimulatory effect on were suppressed knockdown nAChRs using siRNA/α7 nAChR. significantly STAT3 phosphorylation, which attenuated neutralizing activation inhibitor NSC74859 A-inhibited expression. Taken together, our results, for first time, demonstrate that inhibit inflammation through macrophageal subsequent
