摘要: Members of the receptor tyrosine kinase family are frequently implicated in experimental models neoplasia as well human cancer. One best studied signaling systems from this is epidermal growth factor (EGFR). The EGFR widely expressed mammals and has been various stages embryonic development. Recently, knockout mice by targeted disruption exon 1 (Sibilia Wagner 1995) or 2 (Miettinen et al. 1995; Threadgill mouse gene have reported. resulting phenotypes EGFR-/- embryos newborns were very similar. Growth retardation epithelial immaturity found, with severity defects being dependent on genetic background. These results indicate that fundamental importance regulation proliferation differentiation. Experiments transgenic suggest an autocrine mechanism involving could be expected to play a role initiation and/or progression tumors. Transgenic expressing transforming factor-α (TGF-α) reported develop mammary hepatocellular carcinoma pancreatic hyperplasia (Jhappan 1990; Sandgreen 1990).
