摘要: Breast cancer is a major cause of death among women. Although various anticancer drugs have been used in clinics, that are effective against advanced and metastatic breast still lacking great demand. In this study, we found oroxin A, an active component isolated from the herb Oroxylum indicum (L.) Kurz, effectively inhibited growth human cells MDA-MB-231 MCF7 by inducing endoplasmic reticulum (ER) stress-mediated senescence. Oroxin A caused cell cycle arrest at G2/M stage, reorganization microtubules actin cytoskeleton accompanied decrease cellular mitosis. ER-specific probe ER-Tracker Red confocal microscope imaging showed Red-positive increased dosage-dependent manner. addition, population with high β-Gal activity SAHF-positive staining; these data suggest induces ER stress Mechanistic studies led to significant increase intracellular reactive oxygen species levels, promoted expression markers ATF4 GRP78, phosphorylation key stress-response signaling protein p38, resulting Taken together, our indicate exerts its antibreast effects senescence, activating p38 pathway, increasing genes GRP78 levels.
