A caspase-resistant mutant of PKC-|[delta]| protects keratinocytes from UV-induced apoptosis

摘要: Keratinocyte apoptosis induced by UV radiation is a major protective mechanism from skin photocarcinogenesis. The induction of radiation, as well variety genotoxic stimuli, involves the activation PKC-δ caspase-3-mediated cleavage in its hinge region, thus generating constitutively active catalytic fragment. To determine role signaling, we introduced caspase-resistant mutant (D330A) into human keratinocytes retrovirus transduction. Overexpression PKC-δ(D330A) protected UV-induced and enhanced long-term survival. partially prevented release cytochrome c mitochondria loss Mcl-1, key antiapoptotic protein downregulated during apoptosis. Thus, are critical components keratinocytes, inactivation can promote survival exposed to radiation.
