A novel regulatory sequence affecting the constitutive expression of tissue plasminogen activator (tPA) gene in human melanoma (Bowes) cells.

摘要: In order to identify a possible enhancer-like regulatory sequence for the human tissue plasminogen activator (tPA) gene, various DNA segments in 5'-flanking region were ligated chloramphenicol acetyltransferase (CAT) reporter gene driven by tPA promoter, and their CAT enhancing activities measured following transfection melanoma-derived Bowes cells that highly express gene. Major minor detected two adjacent upstream sequences, 160 base pairs (bp) (-2288 -2129) 102 bp (-2390 -2289), respectively, former was subjected further analysis. The activity of 160-bp greatly affected its position orientation constructs also functioned weakly with SV40 promoter. Deletion any small portion from abolished activity, suggesting entire is required activity. This did not show treated inducers phorbol 12 myristate 13-acetate dexamethasone function HeLa or HT1080 under conditions. Taken together, likely be responsible constitutive and/or cell type-specific expression cells.
