Opposite roles of apolipoprotein E in normal brains and in Alzheimer's disease

摘要: We have characterized the interaction between apolipoprotein E (apoE) and amyloid β peptide (Aβ) in soluble fraction of cerebral cortex Alzheimer’s disease (AD) control subjects. Western blot analysis with specific antibodies identified both groups a complex composed full-length apoE Aβ peptides ending at residues 40 42. The apoE–Aβ aggregate is less stable AD brains than controls, when treated anionic detergent SDS. present significantly higher quantity brains, whereas insoluble an inverse correlation has previously been reported. Moreover, subjects bound to more sensitive protease digestion unbound Aβ. Taken together, our results indicate that normal efficiently binds sequesters Aβ, preventing its aggregation. In AD, impaired binding leads critical accumulation facilitating plaque formation.
