Towards proteome scale antibody selections using phage display.

摘要: In vitro antibody generation by panning a large universal gene library with phage display was employed to generate antibodies more than 60 different antigens. Of particular interest comparison of pannings on 20 SH2 domains provided the Structural Genomics Consortium (SGC). Streamlined methods for high throughput developed within 'Antibody Factory' German National Genome Research Network (NGFN) were demonstrated minimise effort and provide reliable robust source antibodies. For domains, in two successive series selections, 2668 clones analysed, resulting 347 primary hits ELISA. Half these further 90 scFv all antigens identified.
