The Uptake, Distribution, and Antitumor Activity of 1-(2-Chloroethyl)-3-cyclohexyl-1-nitrosourea in the Murine Glioma

摘要: The antitumor activity of 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea (CCNU) against the murine ependymoblastoma, glioma 26, and 261 was compared with uptake distribution 14C-labeled drug in ependymoblastoma. With a modified Wilcoxon rank-sum test, it found that CCNU increased life-span tumor animals from 2- to 4-fold, depending on dose used particular type studied. Uptake studies CCNU-14C intracerebral subcutaneous tumors, brain adjacent tumor, distant (normal) indicated parent had relatively constant tissue/plasma ratio levels. Of ether-soluble fractions, greater than 4 accounted for 15% radioactivity. From data, concluded is probably independent preferential between normal tissue. Although few conclusions concerning mode action gliomatous tumors can be made, nevertheless further consideration its use treatment human gliomas seems warranted.
