摘要: In this study, we investigated the suitability of ultrathin and porous polyimide (PI) membrane as a carrier for subretinal transplantation human embryonic stem cell (hESC) -derived retinal pigment epithelial (RPE) cells in rabbits. The vivo effects hESC-RPE were analyzed by suspension injection into Royal College Surgeons (RCS) rats. Rat eyes with electroretinography (ERG) histology. After analyzing surface permeability properties PI, PI transplantations without performed rabbits followed three months fundus photography, ERG, optical coherence tomography (OCT), Animals immunosuppressed cyclosporine entire follow-up time. dystrophic RCS rats, ERG outer nuclear layer (ONL) thickness showed some rescue after injection. Cells positive antigen found clusters under retina 41 days post-injection but not anymore 105 days. rabbits, OCT good placement PI. However, there was loss pigmentation on hESC-RPE-PI over alone, no obvious signs inflammation or atrophy observed. presence hESC-RPE, mononuclear infiltration observed around membranes. well-tolerated space is promising scaffold RPE transplantation. rejection transplanted seems to be major problem given immunosuppression insufficient reduction xenograft induced inflammation.
