摘要: Aims We investigated the sensitivity and specificity of two novel Epidermal growth factor receptor (EGFR) mutation-specific antibodies in detection most common EGFR mutations lung adenocarcinoma. Methods results A total 241 resected adenocarcinoma specimens six post-neoadjuvant gefitinib adenocarcinomas were analysed for mutation using mass spectrometry, fragment analysis direct PCR sequencing platforms. Tissue arrays and/or full sections these cases evaluated immunohistochemistry with (clones SP125 SP111) previously reported 43B2 6B6), specific L858R or 15-nucleotide exon-19 deletion mutations. antibody detected a 76% positive predictive value 73%. SP111 stained deletions 83% 94%. Pretreatment did not affect performance. Full-section immunohistochemical staining heterogeneous mutant proteins expression tumours, revealed non-neoplastic bronchial epithelium adjacent to L858R-carrying carcinomas three 16 (19%) cases. Conclusions Immunohistochemistry mutant-specific may be useful shortening diagnostic time mutations, especially samples low tumour cellularity.
