摘要: Satellite cells isolated from fast tibialis anterior (TA) and slow soleus (SOL) rat muscles were cultivated on matrigel, treated with acidic fibroblast growth factor (aFGF). The following observations made: 1) aFGF-treated cultures exhibited enhanced proliferation as mirrored by a twofold increase in DNA content. 2) Compared to the untreated cultures, myotubes aFGF larger; 3) Using reverse transcriptase polymerase chain reaction (RT-PCR) northern blot analyses, we observed expression of all adult myosin heavy (MHC) isoforms, well myogenin. These findings indicate that, under culture conditions used, has stimulatory effect but also maturation differentiation satellite cells. Furthermore, transcript levels FGF receptor 1 (FGFR1) 4 (FGFR4) bFGF assessed RT-PCR. displayed increased bFGF, suggesting paracrine exogenous aFGF. In this regard, SOL-derived responded more strongly than TA-derived cultures. effects treatment two receptors consisted decrease FGFR1 an FGFR4 mRNA 5-day-old 8-day-old TA similar those 8-day FGF-treated SOL for 8 days higher respective 5 day-treated decreased. This led shift ratio which may explain ability differentiate influence
