摘要: OBJECTIVES To investigate time-course changes in the expression of select genes and extracellular matrix proteins bladder rats with streptozotocin-induced diabetes, so as to examine mechanisms underlying mechanical properties due diabetic cystopathy. METHODS Female Sprague-Dawley were injected streptozotocin (65 mg/kg). Rats that fed 5% sucrose drinking water served diuretic controls, addition normal control rats. At end 2, 4, 8 weeks, total ribonucleic acid (RNA) isolated from detrusor layer bladders was reverse transcribed, then complementary deoxyribonucleic amplified polymerase chain reaction primer sets for type I collagen, III tropoelastin, transforming growth factor beta 1 (TGF-beta-1). Collagen elastin contents quantified commercially available assays. RESULTS Both rat exhibited significantly (P <0.05) lower collagen TGF-beta-1 messenger RNA (mRNA) compared controls at all time points tested.
