摘要: Here we describe the evaluation of 3′-[18F]fluoro-3′-deoxythymidine {[18F]-FLT} as a tracer for positron emission tomography (PET) in murine model B-cell lymphoma and human malignant lymphoma. The line DoHH2 expressed high levels active thymidine kinase 1 (TK-1) key enzyme [18F]-FLT metabolism. Immunostaining confirmed TK-1 derived xenograft tumors SCID/SCID mice. In vitro studies demonstrated time-dependent uptake [18F]-FLT, an efficient phosphorylation to respective monophosphate incorporation into perchloric acid insoluble fraction cells, indicating this DNA. After incubation with [18F]FLT 240 min, 12.5% ± 1.0% radioactivity applied medium was intracellularly trapped cells. Specific accumulation cell clone biodistribution mice bearing DoHH2-derived tumors. percentage injected dose per gram tumor tissue correlated tumor-proliferation index evaluated BrdUrd-labeling experiments. pilot study 11 patients both indolent aggressive lymphoma, suitable comparable [18F]-FDG ability detect lesions by PET scan. Furthermore, found close correlation ( r = 0.95, P < 0.005) standardized values Ki67-labeling biopsies n 10) these patients. These results suggest that represents novel enables imaging proliferation vivo .
