摘要: In the framework of a preliminary investigation on plasma profile cloxacillin after oral administration, simple and rapid LC method was developed for direct determination this compound in human plasma. The on-line sample clean-up carried out using weak anion exchanger (diethylaminoethyl groups) as restricted access material (RAM). effects washing liquid pH, ionic strength addition organic modifier to were studied order obtain an efficient high recovery cloxacillin. separation achieved octadecylsilica stationary phase mobile consisting mixture phosphate buffer (pH 4.0; 25 mM) acetonitrile (72:28, v/v). UV detection performed at 215 nm. most appropriate regression model response function well limit quantitation (LOQ) first selected during pre-validation step. These criteria then assessed formal validation LOQ 50 ng/ml. also validated with respect analyte recovery, precision, trueness, accuracy linearity. Finally, it successfully applied analysis samples obtained from patients having taken dose 500 mg
