摘要: The mycotoxin alternariol (AOH) has been reported to possess genotoxic properties, inducing enhanced levels of DNA damage after only 1 h incubation. In the present study we addressed question whether induction oxidative stress might contribute effects AOH or its naturally occurring monomethylether (AME). dichlorofluorescein (DCF) assay, treatment HT29 cells for formation dichlorofluorescein, indicative ROS formation. total glutathione (tGSH) was transiently decreased. accordance with results DCF and AME proportion transcription factor Nrf2 in nucleus. Concomitantly, Nrf2/ARE-dependent genes γ-glutamylcysteine ligase (γ-GCL) glutathione-S-transferase (GSTA1/2) showed transcript levels. After 24 incubation this effect also reflected on protein level by an increase GST activity. However, spite positive assay activation redox-sensitive Nrf2/ARE-pathway, damage, measured comet addition formamidopyrimidine-DNA-glycosylase (fpg) remained unaffected. Of note, 3 no significant damaging potential detectable, indicating either inactivation compounds repair. summary, mycotoxins were found modulate redox balance but without apparent negative integrity.
