Uptake of IgG in osteosarcoma correlates inversely with interstitial fluid pressure, but not with interstitial constituents.

摘要: The uptake of therapeutic macromolecules in solid tumours is assumed to be hindered by the heterogeneous vascular network, high interstitial fluid pressure, and extracellular matrix. To study impact these factors, we measured fluorochrome-labelled IgG using confocal laser scanning microscopy, pressure 'wick-in-needle' technique, structure stereological analysis, content matrix constituents collagen, sulfated glycosaminoglycans hyaluronan colourimetric assays. microenvironment on factors was studied osteosarcomas implanted either subcutaneously or orthotopically around femur athymic mice. found correlate inversely with tumour volume orthotopic, but not subcutaneous tumours. No correlation between level any constituents. both collagen depended site growth. orthotopic had a higher density than tumours, as surface length were 2-3-fold higher. data indicate that dominant factor controlling tumours; growth important for small vascularization.
