Cinnarizine has an atypical antipsychotic profile in animal models of psychosis

摘要: Cinnarizine, a drug known as calcium channel blocker, is currently used for the treatment of migraine and vertigo. Induction extrapyramidal signs by cinnarizine has been reported in elderly, which related to its moderate antagonistic properties at dopamine D2 receptors, resembling mechanism action most antipsychotic drugs. Despite this effect, never tested putative drug. Here we evaluate potential effect two pharmacological models psychosis, namely amphetamine- MK-801-induced hyperlocomotion, well ability induce catalepsy. Cinnarizine significantly counteracted MK-801 (0.25 mg/kg) amphetamine (5mg/kg) locomotor effects doses low 20mg/kg, having no incremental 60 or 180 mg/kg. Regarding side-effects, induced catalepsy mice effective dose 20 mg/kg, inducing only mild Based on these results antagonist suggest that it with an atypical profile should be evaluated clinically.
