摘要: Angiogenesis is regulated by the highly coordinated function of various proteins with pro- and antiangiogenic functions. Proangiogenic factors include vascular endothelial growth factor (VEGF), fibroblast factor, platelet-derived insulin-like transforming angiopoietins, several chemokines; thrombospondin-1, angiostatin, endostatin. Matrix metalloproteinases display a dual role in development. Notch signaling affects remodeling primary network uniformly sized vessels into functionally morphologically distinct arteries, veins, capillaries. Tumors, described as 'wounds that never heal', lose appropriate balance among these factors. Although VEGF-targeted therapies are showing promise, new angiogenesis targets needed to make additional gains. Here, we highlight recent advances our understanding regulation tumor discuss potential molecular targeting therapeutic approach.
