Absence of Methylthioadenosine Phosphorylase in Human Gliomas

摘要: All normal mammalian tissues contain methylthioadenosine phosphorylase, which plays a role in the recycling of purines and methionine consumed during polyamine synthesis. A complete deficiency phosphorylase has been reported some human leukemias lymphomas few solid tumors. The exact incidence enzyme among fresh tumor specimens difficult to establish because measurement catalytic activity is laborious requires carefully preserved specimens. We have generated two antibodies against used them develop simple immunoblot assay for enzyme. Specifically, studies showed that all cells with catalytically active had 32-kDa band reacted anti-enzyme antibodies. In reciprocal manner, malignant cell lines were naturally deficient lacked detectable immunoreactive protein. immunoassay was analyze gliomas. Seventy-five % (9 12) gliomas completely deficient. This common metabolic difference between most potential target tumor-specific chemotherapy.
