Temporal heterogeneity in single-cell gene expression and mechanical properties during adipogenic differentiation

摘要: Adipose-derived stem/stromal cells (ASCs) respond heterogeneously when exposed to lineage-specific induction medium. Variable responses at the single-cell level can be observed in production of metabolites, expression mRNA transcripts, and adoption mechanical phenotypes. Understanding relationship between biological characteristics for individual ASCs is crucial interpreting how cellular heterogeneity affects differentiation process. The goal current study was monitor gene peroxisome proliferator receptor gamma (PPARG) adipogenically differentiating ASC populations over two weeks, while also characterizing expression-associated properties using atomic force microscopy (AFM). Results showed that did not change significantly time either adipogenic or control medium; however, expressing PPARG exhibited greater compliance fluidity compared those lacking both media environments. percent PPARG+ samples increased but stayed relatively constant controls.
