摘要: Dahl SS rats exhibit greater levels of renal medullary oxidative stress and lower fumarase activities. Fumarase insufficiencies can increase reactive oxygen species (ROS), the mechanism which, however, is not clear. A proteomic analysis indicated knockdown in HK-2 cells resulted changes expression or activity NADPH oxidase, mitochondrial respiratory chain complex I III, ATP synthase subunits, α-oxoglutarate dehydrogenase, all which are sites ROS formation. Meantime, activities key antioxidant enzymes such as G6PD, 6PGD, GR, GPx GST increased significantly too. The apparent activation defense appeared insufficient glutathione precursors, GSH/GSSG ratio were decreased. exhibited redox metabolism similar to with knockdown. Supplementation fumarate malate, substrate product fumarase, decreased, respectively, blood pressure H2O2 MDA kidney tissues rats. These results indicate cause a wide range at several production mechanisms.
