Brain tumor development in rats is associated with changes in central nervous system cytokine and neuropeptide systems

摘要: Cytokines have roles in tumor biology and induce neurological manifestations. produced response to a brain may generate manifestations via paracrine action. We investigated cytokine modulation an vivo model with behavioral, morphological, molecular approaches. Rat C6 glioma cells were implanted into the third cerebral ventricle of Wistar rats, their behavior was monitored, development intracranial astrocytic origin confirmed by histology positive immunostaining for vimentin, S-100 protein, glial fibrillary acidic protein. Sensitive specific RNase protection assays used analyze messenger RNA (mRNA) regions from anorexic tumor-bearing animals. Brain formation associated significant increased levels interleukin (IL)-1β, IL-1 receptor antagonist, type I, necrosis factor (TNF)-α, transforming growth (TGF)-β1 mRNAs cerebellum, hippocampus, hypothalamus. accessory proteins I II cerebellum also examined hypothalamic feeding-associated components: neuropeptide Y proopiomelanocortin down-regulated, glycoprotein 130 mRNA up-regulated, leptin (OB-R) unchanged. These dissimilar profiles expression suggest specificity tumor-induced transcriptional changes. The data implicate cytokines as important factors tumor-host interactions vivo. show that cell-induced can be behavioral-molecular study action during host biochemical physiological responses development. Paracrine seem pivotal because observed various regions. results changes progression are involved tumor-associated neuropsychiatrical
