摘要: The human high m.w.-melanoma-associated Ag (HMW-MAA) is an attractive target for the immunotherapy of melanoma, due to its relatively expression in a percentage melanoma lesions and restricted distribution normal tissues. Active immunization with HMW-MAA mimics has been previously shown induce HMW-MAA-specific, T cell-dependent Ab response associated apparent clinically beneficial effect advanced patients. Although cells play important role controlling tumor growth, only limited information available date about induction HMW-MAA-specific CTL. In this report, we show that HLA-A2/Kb transgenic mice cDNA-transfected syngeneic dendritic elicited CD8+ CTL specific peptides HLA-A2 Ag-binding motifs. lysed HLA-A2+HMW-MAA+ vitro, mouse pulsed HMW-MAA-derived vitro vivo. could be generated absence CD4+ cell help, harnessing help noncognate Ag-specific manner polyclonal activator staphylococcal enterotoxin A augmented response. These results imply cell-based immunization, combination represents effective strategy implement targeting patients HMW-MAA-bearing tumors.
