摘要: Introduction: Glioblastoma multiforme (GBM) is one of the deadliest forms brain cancer, and affects more than 18,000 new cases each year in United States alone. The current standard treatment for GBM includes surgical removal tumor, along with radiation chemotherapy. Despite these treatments, recurrence extremely common, once it recurs, life expectancy measured weeks or months. One reasons deadly nature recurrent thought to be selection therapy-resistant tumor cells. In this project, we sought characterize molecular changes specimens compared primary from same subjects. Methods: Whole-genome DNA microarrays were used identify genes changed mRNA expression seven samples Real-time quantitative RT-PCR was an attempt validate seen by microarray 18 interest chosen screen. Results: experiments identified several dozen transcripts evidence significant differences expression. From genes, chose PCR validation. Overall, validated findings quite well. There a very high correlation magnitude (Pearson’s R = 0.852, P < 0.001). Individually, 13 showed statistically versus pairs. Of 5 that did not at P<0.05 level, 4 trends direction predicted microarray, while 1 gene not. Conclusion: Real time has proven useful validating GBMs could have important clinical applications.
