摘要: Colorectal cancer is the second leading cause of death in United States. The nihilism that previously often characterized treatment patients with this disease has been replaced by a measure excitement, given recent therapeutic advances. These advances have stimulated part through identification cellular processes characteristic colorectal cancers permit targeting favorable index. Inhibition epidermal growth factor receptor clinic provided proof principle interruption signal transduction cascades potential. This experience also taught us resistance to such rationally developed targeted strategies common. In article, we review role cancer, introduce promising molecular targets, and outline approaches under development. We will describe barriers success, highlight paradigms facilitate rapid successful evaluation new agents, clinical context (by virtue successes) become somewhat crowded playing field.
