Expression cloning of human EGF receptor complementary DNA: gene amplification and three related messenger RNA products in A431 cells

摘要: In order to further define the mechanisms by which polypeptide growth factors regulate gene transcription and cellular growth, expression cloning techniques were used select human epidermal factor (EGF) receptor complementary DNA clones. The EGF 3' coding domain shows striking homology transforming product of avian erythroblastosis virus (v-erbB). Over-expression receptors in A431 cell lines correlates with increased mRNA levels amplification (up 110 times) apparently singular gene. There appear be three cytoplasmic polyadenylated RNA products cells, one contains only 5' (EGF binding domain) sequences is postulated encode secreted receptor-related protein.
