摘要: Two human breast cancer cell lines of differing invasive and metastatic potential, MDA-MB-435 MCF7, were examined using subtractive suppression hybridization in a search for any genes associated with metastasis. Of the 17 cDNAs identified as being differentially expressed genes, it was determined that syntenin overexpressed cells. Expression analysis showed expression level well correlated potential various gastric lines. Moreover, tumor tissues exhibited much higher mRNA than their normal counterparts. Syntenin-transfected MCF7 cells migrated more actively, an increased invasion rate relative to vector-transfectants or parental vitro, without evidencing effect on adhesion fibronectin, type I collagen laminin. Similarly, forced line Az521 its migratory vitro. Syntenin-expressing appearance numerous surface extensions pseudopodia formation I, suggesting may be involved signaling cascade actin-reorganization. Mutation study suggested PDZ2 domain could essential role stimulatory migration. This is first demonstration syntenin, PDZ motif-containing protein, can during progression function metastasis-inducing gene.
