In Vitro and in Vivo Anti-Inflammatory Effect of a Biotechnologically Modified Borage Seed Extract: Evidence for Lipid Pro-Resolving Mediators’ Implication in the Enhancement of Psoriatic and Atopic Dermatitis Lesions

摘要: Aim: Resolvins, maresins and lipoxins are lipid mediators issued from essential polyunsaturated fatty acids which the first anti-inflammatory pro-resolving signals identified during resolution phase of inflammation. As borage oil and/or borage seed extracts have shown beneficial action in treatment atopic dermatitis or eczema human canine, we modified a component by using biotechnology order to get compound structurally related polyunsaturated fatty acid, studied its ability reduce inflammation production through generation resolvins, and/or lipoxins. Additionally, we have demonstrated potent effect this new which consists aminopropanediol amides, through an vivo study concerning subjects suffering from psoriasis or dermatitis. Study Design/Methods: For vitro study, was induced in co-cultures dendritic cells normal keratinocytes addition of PMA calcium ionophore A23187. Ability our aminopropanediol amides increase resolvin D2, maresin 1 A4 B4 synthesis then measured. Pro-inflammatory cytokines (IL-1β, IL-6, IL-8) PGE2 productions were also quantified. 36 suffering from psoriasis dermatitis used twice day 30 days, formulation containing amides. Before beginning the study after days’ treatment, severity dermatitis was evaluated using PGA SCORAD scoring scales, respectively. Results: Borage amides able significantly the resolvin synthesis. Concomitantly, they were also inhibit production IL-1β, IL-6, IL-8 induced ionophore A23187 co-culture model used. Introduced formulation, significantly reduced clinical manifestations Conclusion: Our clearly showed activity borage seed emphasized putative role pro-resolving lipid dermatitis, other inflammation-induced skin diseases.
